Nadal beat Italian Andreas Seppi in straight sets 6-1 6-3, to qualify for the quarterfinals of the Monte Carlo Masters.
The Spaniard will next play against his compatriot David Ferrer in the next round of the competition, after he defeated Bulgarian world number 14 Grigor Dimitrov 6-4 6-2.
But Nadal played down the importance of the 300th win, saying that his focus is on the next game.
"I did what I had to," said Nadal. "But playing David (Ferrer) is never easy. If you are not at the top of your game, you will lose. I'm just glad to be in a quarter-final."
Meanwhile Novak Djokovic eased his way into the last eight of the competition, effortlessly seeing off the challenge of Pablo Carreno 6-0 6-1.
Also Roger Federer qualified for the last eight of the Monte Carlo tournament,in a 6-4 6-1 win over Lukas Rosol.
