Wozniacki for once stepped up her game to beat a top player after giving up a set in her 6-4 2-6 6-2 win over Sharapova at Flushing Meadow.
Sharapova with the loss,has become the latest highest ranked player to be eliminated,with Serena Willams the only player out of the top five still left in the competition.
"It means so much to me - it's been a bit up and down for me this year," said Wozniacki.
"It's a new year and I've gotten older, and hopefully a little wiser. I feel good. I feel comfortable when I play out there.
She will next play against Sara Errani in the next round of the tournament.
Meanwhile in the Men's draw,Roger Federer came from behind to beat Marcel Granollers in four sets game.
The 17 Grand Slam winner beat Granollers 4-6 6-1 6-1 6-1 to qualify for the next round of the competition.
Fourth seed David Ferrer was the first top-10 casualty in the men's draw, losing to Gilles Simon 6-3 3-6 6-1 6-3.
